There is Standard and Hell mode. In Standard, you can get hit 3 times before you die (falling down a pit is still instant death). In Hell, one hit and you're dead.
You will get something like half a second invincibility after being hit in Standard. You can also use spinning and dashing for temporary invincibility.
If you play on Hell mode it is very possible for you to go through all 1,000 lives. On Standard, if you're good, you'll probably make it through before that.
The only way to get lives back is to use an option to start over from the beginning. You'll keep all earned records and data, but start the story over. You'll have to do this anyway if you want to see and collect absolutely everything.

Thanks! Also, how do you get the infinity symbol that I saw in one of the screenshots, and how does that factor into gameplay?

If I'm thinking of the right screenshot, that's just the tutorial stages where you get infinite prinnies to practice and die to your heart's content.
